The expression of detoxification genes in two maize cultivars by interaction of isoxadifen-ethyl and nicosulfuron

Herbicide safeners protect crop plants from herbicide phytotoxicity, but an understanding of their molecular mechanisms is still lacking. We investigated the effects of the safener isoxadifen-ethyl and/or nicosulfuron on the expression of 10 genes, 8 glutathione transferases (GSTs), 1 glutathione transporter and 1 multidrug resistance protein gene in two maize cultivars. Nicosulfuron and isoxadifen-ethyl induce different detoxification enzyme genes. The expression analyses of the 10 genes revealed that most were expressed much higher in ‘Zhengdan958’ than those in ‘Zhenghuangnuo No.2’, both in control and in isoxadifen-ethyl- and/or nicosulfuron-treated plants. The expression levels of ZmGSTIV, ZmGST6, ZmGST31 and ZmMRP1 in two maize cultivars were up-regulated by isoxadifen-ethyl only, or in combination with nicosulfuron, whereas nicosulfuron down-regulated the expression of eight genes. Thus, ZmGST1V, ZmGST6, ZmGST31 and ZmMRP1 could be considered safener-responsive and may be the core genes responsible for isoxadifen-ethyl increasing the tolerance of maize to nicosulfuron.

SYNTHESIS, CRYSTAL STRUCTURE AND BIOACTIVITY OF N-PHENOXYACETYL-2-SUBSTITUTED-1,3-OXAZOLIDINES

A series of novel N-phenoxyacetyl-1,3-oxazolidine derivatives were synthesized by the cyclization and acylation with ethanolamine, ketone and phenoxyacetyl chloride as the starting materials. Design, synthesis, and anticancer evaluation of some novel thiourea, carbamimidothioic acid, oxazole, oxazolidine, and 2-amino-1-phenylpropyl-2-chloroacetate derived from L-norephedrine

A novel series of thiourea, carbamimidothioic acid, 4, 5-dihydrooxazole-2-thiol, oxazolidine-2thine, and 2-amino-1-phenylpropyl-2-chloroacetate derivatives was designed and synthesized using 2-amino-1-phenylpropan-1-ol (L-norephedrine) as a strategic starting material. The structures of the newly synthesized compounds were established by elemental analyses, IR, and H-1 NMR and C-13 NMR spectral data. The compounds were evaluated for their in vitro anticancer activity against various cancer cell lines. The corresponding acetamide, carbamimidothioic acid, and 2-2-amino-1-phenylpropyl-2-chloroacetate derivatives showed almost the same activity as the standard drug doxorubicin against human breast cancer cell line (MCF-7). Acid-base disorders disrupt proper cellular functions, which are associated with diverse diseases. Development of highly sensitive pH probes being capable of detecting and monitoring the minor changes of pH environment in living systems is of considerable interest to diagnose disease as well as investigate biochemical processes in vivo. We report herein two novel high-resolution ratiometric two-photon (TP) fluorescent probes, namely, PSIOH and PSIBOH derived from carbazole-oxazolidine pi-conjugated system for effective sensing and monitoring acid pH in a biological system. Remarkably, PSIOH exhibited the largest emission shift of similar to 169 nm from 435 to 604 nm upon pH changing from basic to acidic with an ideal pK(a) value of 6.6 within a linear pH variation range of 6.2-7.0, which is highly desirable for high-resolution tracking and imaging the minor fluctuation of pH in live cells and tissues. PSIOH also exhibits high pH sensitivity, excellent photostability, and reversibility as well as low cytotoxicity. More importantly, this probe was successfully applied to (i) sense and visualize the pH alteration in HeLa cells caused by various types of exogenous stimulation and (ii) detect and differentiate cancer and tumors in liver tissues and a mouse model, realizing its practical in vitro and in vivo applications.
