Spiramine C-D, the atisine-type diterpenoid alkaloids isolated from the Chinese herbal medicine Spiraea japonica complex, are shown to have anti-inflammatory effects in vitro. In this study, we report that spiramine derivatives of spiramine C-D bearing alpha,beta-unsaturated ketone induce apoptosis of Bax (/) /Bak (/) MEFs cell, which is positively corresponding their cytotoxicity of tumor cell lines including multidrug resistance MCF-7/ADR. The results indicated that oxazolidine ring is necessary, and derivatives bearing double ‘Michael reaction acceptor’ group would significantly increased activities both of inducing apoptosis of Bax (/) /Bak (/) cells and cytotoxicity of tumor cells. The result indicated that spiramine derivative with alpha,beta-unsaturated ketone group is a new anti-cancer agent with a capability of inducing apoptosis of cancer cells in Bax(-/-)/Bak(-/-) independent manner. A Simple and Efficient Approach for the Synthesis of 1,3-Oxazolidines from beta-Amino Alcohols Using Grinding Technique

An environment-friendly and efficient method for synthesis of 1, 3-oxazolidines from beta-amino alcohols has been developed using paraformaldehyde in the presence of 10 mol% alkali as a green reaction medium. The method is concise, simple, transition metal free, and high yielding at room temperature with wide substrate scope and functional group compatibility. The optimized method can be used for the synthesis of 1, 3-oxazolidines intermediates, which are used as prodrug moieties to improve therapeutic potential of the parent drug. Structure and formation of the synthesized compounds has been characterized and confirmed through conventional spectroscopic techniques, such as H-1 nuclear magnetic resonance (NMR), C-13 NMR, and mass spectroscopy.

Metal-Free Synthesis of Oxazolidine-2,4-diones and 3,3-Disubstituted Oxindoles via ICl-Induced Cyclization

A metal-free method for the construction of oxazolidine-2,4-diones and oxindoles was discussed. Using iodine monochloride (ICl) as both the reaction promoter and iodide source, the iodolactonization of N-Boc acrylamides proceeded readily and provided the corresponding iodo oxazolidine-2,4-diones and oxazolidin-2-ones in good isolated yields. Synthesis of chiral palladium oxazolidine and imine complexes: Investigation the oxazolidine-imine conversion by DFT method

Reaction of 2-pyridinecarboxaldehyde with L-Alaninol (as a chiral amine) or 2-amino-2-methyl-1-propanol (AMP) in the presence of PdCl2 produced the new and attractive palladium(II) complexes which were synthesized by in situ method. The presence of an extra methyl group in AMP other than L-Alaninol induced the different chelating mode and, therefore, several structurally different palladium complexes obtained. Correspondingly, the chiral amine with PdCl2 produced the imine product, (Iminol (1)), and an oxazolidine compound (Imizol (1′)) as minor product. Also, the use of AMP in formation of palladium complexes led to the synthesis of imine (Ampynol (2)) and oxazolidine (Ampyzol (3)) complexes. Besides utilizing of crystallization technique for the separation of the isomers, elemental analysis (CHN), FT-IR, H-1 and C-13 NMR spectroscopies were used for characterization of the synthesized compounds. In addition, structures of palladium complexes Iminol and Ampyzol were identified by single crystal X-ray diffraction method and investigation of the equilibrium between imine and oxazolidine complexes were carried out by DFT method. The in vitro studies revealed that the compounds have considerable cytotoxicity against human MCF-7 and HeLa cancer cell lines. (C) 2018 Elsevier Ltd. All rights reserved.

Preparation and Enantioseparation of a New Click Derived beta-Cyclodextrin Chiral Stationary Phase

A new cyclodextrin-derived chiral stationary phase (denoted as CDA-CSP) was synthesized by immobilizing mono(6(A)-azido-6(A)-deoxy)-per(p-chlorophenyl carbamoylated) beta-cyclodextrin derivative to alkynyl modified silica via click chemistry. This newly prepared CSP shows good enantioseparation performance for six chiral compounds (1-6), such as 4-phenyl-oxazolidine-2-thione, two kinds of aryl alcohols, substituted flavonoids and benzoin, in which baseline separation of Analytes 1-4 was achieved under the experimental conditions. The effects of column temperature, mobile phase pH and content of methanol on the enantioseparation characteristics of CDA-CSP were investigated in detail. Retention factor and resolution for Compound 3 gradually reduced with an increase of column temperature, and a good linear relationship was shown between napierian logarithm of selectivity factor and reciprocal of column temperature. In the pH range from 3.56 to 5.50, a change in pH hardly affected the resolution of Analyte 2. In addition, increasing methanol in the mobile phase resulted in rapid eluting of the analytes from the column in reversed-phase mode. The retention factors for Analytes 1 and 3 significantly decreased and their resolution showed different trends.

Protection of abasic sites during DNA replication by a stable thiazolidine protein-DNA cross-link

A basic (AP) sites are one of the most common DNA lesions that block replicative polymerases. 5-hydroxymethylcytosine binding, embryonic stem cell-specific protein (HMCES) recognizes and processes these lesions in the context of single-stranded DNA (ssDNA). A HMCES DNA-protein cross-link (DPC) intermediate is thought to shield the AP site from endonucleases and error-prone polymerases. The highly evolutionarily conserved SOS-response associated peptidase (SRAP) domain of HMCES and its Escherichia coli ortholog YedK mediate lesion recognition. Here we uncover the basis of AP site protection by SRAP domains from a crystal structure of the YedK DPC. YedK forms a stable thiazolidine linkage between a ring-opened AP site and the alpha-amino and sulfhydryl substituents of its amino-terminal cysteine residue. The thiazolidine linkage explains the remarkable stability of the HMCES DPC, its resistance to strand cleavage and the proteolysis requirement for resolution. Furthermore, its structure reveals that HMCES has specificity for AP sites in ssDNA at junctions found when replicative polymerases encounter the AP lesion.
